Unfortunately there is no option to accent any notes in Hookpad. All melody notes have the same velocity.
The only way I can think to achieve something like this would be to use two voices, voice1 and voice2 for example, select the same lead instrument for both and turn down the volume of one of the instruments. Now you can write louder and quieter notes with the same sound. But this is more a hack than a solution might be a bit complicated.
